Sonidegib (INN), also known as LDE225 and marketed as Odomzo,[2] is a Hedgehog signaling pathway inhibitor (via smoothened antagonism) being developed as an anticancer agent by Novartis.[3][4]
Approvals and indications
It was approved by the FDA for treating basal cell carcinoma in July 2015[5][2] and is awaiting approval in the EU. In the EU, the agent has received positive opinion from the CHMP for approval.
Investigations
It has been investigated as a potential treatment for:
- Pancreatic cancer[6][7][8][9]
- Breast cancer[10][11]
- Basal cell carcinoma of the skin[12][13][14]
- Small cell lung cancer[15]
- Medulloblastoma[16][17]
- Advanced solid tumours (including ovarian, breast, pancreatic, stomach, oesophageal cancers and glioblastoma multiforme)[18][19][20]
- Acute leukaemia[21]
- Chronic myeloid leukaemia[22]
- Myelofibrosis and essential thrombocythaemia[23]
It has demonstrated significant efficacy against melanoma in vitro and in vivo.[24] It also demonstrated efficacy in a mouse model of pancreatic cancer.[25]
